Inhibitory activity of CXCR4 attenuates intervertebral disc degeneration by regulating TXNIP/NLRP3 expression

چکیده
Purpose: To explore the inhibitory effect and mechanism of action CXCR4 on intervertebral disc degeneration. Methods: The expression was assessed in different degrees degenerate tissues. Nucleus pulposus (NP) cells-induced degeneration were obtained using interleukin 1 beta (IL-1β) stimulation. inhibitor, AMD 3100, employed to resist function CXCR4, order determine whether inhibition acted by targeting TXNIP/NLRP3 axis. Results: Expression increased severely degenerated IL-1β activated promoted progression degeneration, but AMD3100 treatment reversed effects IL-1β. Moreover, TXNIP NLRP3 significantly suppressed stimuli. Conclusion: results demonstrate that prevents NP cells from degradation regulating signaling pathway, thus indicating this approach could be effective management degeneration.

Degeneration of the intervertebral disc
The intervertebral disc is a cartilaginous structure that resembles articular cartilage in its biochemistry, but morphologically it is clearly different. It shows degenerative and ageing changes earlier than does any other connective tissue in the body. It is believed to be important clinically because there is an association of disc degeneration with back pain. Current treatments are predomina...
متن کاملIntervertebral Disc Dynamics and Degeneration
Degenerative disc disease is a serious health problem worldwide, whose etiological basis – mechanical stimulus, biochemical changes, or natural aging – is poorly understood. While the disc’s known function is that of dynamic shock absorber, its dynamic response has not been fully characterized throughout the disc health spectrum (from healthy to degenerate). Therefore, this study aimed to corre...

Transplantation of nucleus pulposus cells (NPCs) into the intervertebral disc (IVD) has been demonstrated to be an effective treatment of degenerative disc disease (DDD). However, the underlying mechanisms have remained to be sufficiently elucidated. The aim of the present study was to explore the potential cell migration and anti‑apoptosis efficacy of NPCs in the treatment of DDD. NPCs culture...
متن کاملGene expression profile analysis of human intervertebral disc degeneration
In this study, we used microarray analysis to investigate the biogenesis and progression of intervertebral disc degeneration. The gene expression profiles of 37 disc tissue samples obtained from patients with herniated discs and degenerative disc disease collected by the National Cancer Institute Cooperative Tissue Network were analyzed. Differentially expressed genes between more and less dege...
